- High background → optimise block, dilute antibody, wash more
- False negative → check fixation, retrieval, antibody, controls
- Weak signal → stronger retrieval, longer incubation
- Edge artifact → keep sections covered, avoid drying
- Control fails → re-run batch; do not report
- Always run positive + negative controls
