Soft paraffin block (won't section cleanly)
Also known as: mushy block, block won't hold an edge
Block face smears or the tissue drags; sections are thick, torn or won't form.
Where it usually appears: Under-processed tissue, wax contaminated with clearing agent, or block not cooled.
- Incomplete dehydration/clearing so wax did not fully infiltrate
- Embedding wax contaminated with xylene/clearant
- Block not chilled before sectioning
- Wax melting point too low for the room
- Chill the block thoroughly on a cold plate/ice
- Check wax bath temperature and cleanliness
- If under-processed, reprocess the tissue
- Reprocess under-infiltrated tissue through fresh reagents
- Change contaminated embedding wax
- Maintain reagent rotation/change schedules
- Keep the last wax baths free of clearant carry-over
- Cool blocks fully before cutting
