Out-of-focus regions on whole-slide images

What it looks like

Regions of the whole-slide image appear blurred or lacking detail, making it difficult to assess cellular structures.

Where it usually appears: Typically observed in areas of the slide that are not properly focused during scanning.

Likely causes
  • Inadequate focus during scanning
  • Slide thickness variation
  • Incorrect Z-axis calibration
  • Dirt or damage on the objective lens
Immediate checks
  • Verify the focus setting of the scanner
  • Inspect the slide for physical damage or uneven thickness
Corrective action
  • Re-scan the slide with adjusted focus
  • Clean the objective lens thoroughly
Preventive action
  • Regular maintenance of the scanning equipment
  • Ensure slides are prepared with consistent thickness
