Tissue Processing SOP
Purpose
To dehydrate, clear and infiltrate tissue with paraffin for sectioning.
Scope
Automated tissue processor operation.
Responsibilities
- Processing technician: run and monitor cycles
- Lab manager: reagent rotation
Materials & equipment
- Graded alcohols, clearing agent, paraffin wax
- Tissue processor
Procedure
- Select the schedule appropriate to specimen type (short for biopsies, longer for fatty/dense tissue).
- Confirm reagent levels and rotation status before starting.
- Load baskets and start the validated cycle.
- On completion, transfer to the embedding station.
- Record cycle, schedule and any alarms.

- Processor run log
- Reagent change log
Safety
- Reagents flammable/toxic — ensure ventilation and PPE.
