Special Stains (General) SOP
Purpose
To outline the procedures for performing special stains in histopathology to ensure consistent and accurate results.
Scope
This SOP applies to all laboratory personnel involved in staining procedures.
Responsibilities
- Laboratory Technologist: Perform special stains according to the SOP.
- Quality Manager: Ensure compliance with SOP and review periodically.
- Laboratory Supervisor: Train staff on special staining techniques.
Materials & equipment
- Staining reagents
- Microscope slides
- Coverslips
- Staining racks
- Distilled water
- Pipettes
Procedure
- Gather all necessary materials and reagents before starting the staining process.
- Label all slides clearly with patient or sample identifiers.
- Prepare the staining reagents according to the manufacturer's instructions.
- Deparaffinize and rehydrate the tissue sections as required.
- Apply the special stain to the tissue section as per the specific protocol.
- Rinse the slides with distilled water after staining.
- Counterstain if required and rinse again.
- Dehydrate the slides through graded alcohols if needed.
- Clear the slides in xylene or appropriate clearing agent.
- Apply coverslips using mounting medium.
- Examine the stained slides under a microscope and document findings.
Records
- Staining logbook to record date, technician, and staining results.
- Quality control records for staining reagents.
Safety
- Wear appropriate personal protective equipment (PPE) including gloves and lab coat.
- Handle all reagents according to their safety data sheets (SDS).
- Dispose of hazardous waste according to laboratory waste disposal guidelines.
