Reticulin Staining SOP
Purpose
To provide a standardized method for performing reticulin staining in histopathology to visualize reticular fibers.
Scope
This SOP applies to all laboratory personnel involved in staining procedures.
Responsibilities
- Laboratory Technician: Perform staining according to SOP
- Quality Manager: Ensure compliance and review SOP
- Pathologist: Interpret stained slides
Materials & equipment
- Reticulin stain kit
- Microscope slides
- Cover slips
- Distilled water
- Ethanol
- Mounting medium
Procedure
- Prepare tissue sections on microscope slides.
- Dehydrate slides through graded ethanol series.
- Incubate slides in reticulin stain solution for specified time.
- Rinse slides in distilled water to remove excess stain.
- Counterstain with appropriate counterstain if required.
- Dehydrate slides again through graded ethanol series.
- Clear slides in xylene or clearing agent.
- Apply mounting medium and cover slips.
- Allow slides to dry before microscopic examination.
- Document staining details in laboratory records.

Safety
- Wear appropriate personal protective equipment (PPE)
- Handle chemicals according to safety data sheets (SDS)
- Dispose of waste according to laboratory protocols
