PAS Staining SOP
Purpose
To provide a standardized method for performing Periodic Acid-Schiff (PAS) staining to identify polysaccharides in tissue samples.
Scope
This SOP applies to all histopathology laboratory personnel involved in staining procedures.
Responsibilities
- Technician: Prepare and perform PAS staining according to this SOP.
- Quality Manager: Ensure compliance with the SOP and maintain records.
Materials & equipment
- Periodic Acid solution
- Schiff's reagent
- Distilled water
- Microscope slides
- Cover slips
- Staining jars
Procedure
- Prepare tissue sections on microscope slides.
- Deparaffinize and hydrate sections through graded alcohols to water.
- Immerse slides in Periodic Acid solution for 5-10 minutes.
- Rinse slides in distilled water for 1-2 minutes.
- Apply Schiff's reagent to the sections for 15-30 minutes in the dark.
- Rinse slides in running tap water for 5 minutes.
- Counterstain with hematoxylin for 1-2 minutes if desired.
- Rinse slides in distilled water.
- Dehydrate through graded alcohols and clear in xylene.
- Mount with a cover slip and label the slides.
Records
- Staining log indicating date, operator, and samples stained.
- Quality control records for reagents used.
Safety
- Wear appropriate personal protective equipment (PPE) including gloves and lab coat.
- Handle all chemicals according to their safety data sheets (SDS).
