Pap Smear Staining SOP
Purpose
To outline the standardized procedure for staining Pap smears in cytology to ensure consistent and accurate results.
Scope
This SOP applies to all personnel involved in the staining of Pap smears in the laboratory.
Responsibilities
- Laboratory personnel: Perform staining according to this SOP
- Quality manager: Ensure compliance and training on the SOP
Materials & equipment
- Pap smear slides
- Staining reagents (e.g., Papanicolaou stain)
- Distilled water
- Mounting medium
- Glass coverslips
- Staining jars
Procedure
- Collect Pap smear slides for staining.
- Prepare staining reagents according to manufacturer instructions.
- Dehydrate slides through graded alcohols (70%, 95%, 100%).
- Clear slides in xylene or a suitable clearing agent.
- Stain slides with Papanicolaou stain for specified time.
- Rinse slides in distilled water to remove excess stain.
- Dehydrate slides again through graded alcohols.
- Clear slides in xylene or a suitable clearing agent.
- Apply mounting medium and coverslips to slides.
- Allow slides to dry before storage.
- Label slides appropriately for identification.
Records
- Staining log: Document date, technician, and batch number of slides stained.
- Quality control records: Document any QC checks performed on staining reagents.
Safety
- Wear appropriate personal protective equipment (PPE) including gloves and lab coat.
- Handle all reagents according to their safety data sheets (SDS).
- Dispose of waste materials according to hazardous waste guidelines.
