Microtomy SOP
Purpose
To cut and mount high-quality paraffin sections.
Scope
Rotary microtome operation.
Responsibilities
- Microtomy technician
Materials & equipment
- Rotary microtome, blades
- Floatation bath, slides, cold plate
Procedure
- Chill the block, clamp firmly, and face the block.
- Cut sections at the standard thickness (typically 3–5 µm).
- Float on the water bath (~8–10°C below wax MP) and mount on labelled slides.
- Confirm slide identity matches the block; dry on the slide warmer.
- Change blades on schedule; keep sharps in a designated container.

Safety
- Blade safety — lock/guard when not cutting; sharps disposal.
