Liquid-Based Cytology SOP

Purpose

To outline the standardized procedures for performing liquid-based cytology to ensure accurate and reliable results.

Scope

This SOP applies to all personnel involved in liquid-based cytology in the laboratory.

Responsibilities

  • Cytotechnologist: Perform liquid-based cytology procedures and ensure quality control.
  • Laboratory Manager: Oversee compliance with this SOP and staff training.
  • Quality Manager: Review and update the SOP as necessary.

Materials & equipment

  • Cytology collection device
  • Preservative solution
  • Centrifuge
  • Microscope
  • Slide preparation materials
  • Labeling materials

Procedure

  1. Collect specimen using the cytology collection device according to manufacturer instructions.
  2. Immediately place the collected specimen into the preservative solution.
  3. Mix the specimen thoroughly to ensure even distribution of cells.
  4. Centrifuge the specimen at the specified speed and time as per protocol.
  5. Decant the supernatant carefully without disturbing the cell pellet.
  6. Resuspend the cell pellet in an appropriate volume of preservative solution.
  7. Prepare slides by placing the resuspended cell sample onto the slide and spreading it evenly.
  8. Label the slides with appropriate patient identification and date.
  9. Allow the slides to dry completely before staining.
  10. Stain the slides using the standardized staining protocol.
  11. Examine the slides under a microscope and record findings.
  12. Document all results in the laboratory information system.
