Purpose
To outline the standardized procedures for performing liquid-based cytology to ensure accurate and reliable results.
Scope
This SOP applies to all personnel involved in liquid-based cytology in the laboratory.
Responsibilities
- Cytotechnologist: Perform liquid-based cytology procedures and ensure quality control.
- Laboratory Manager: Oversee compliance with this SOP and staff training.
- Quality Manager: Review and update the SOP as necessary.

- Cytology collection device
- Preservative solution
- Centrifuge
- Microscope
- Slide preparation materials

Procedure
- Collect specimen using the cytology collection device according to manufacturer instructions.
- Immediately place the collected specimen into the preservative solution.
- Mix the specimen thoroughly to ensure even distribution of cells.
- Centrifuge the specimen at the specified speed and time as per protocol.
- Decant the supernatant carefully without disturbing the cell pellet.
- Resuspend the cell pellet in an appropriate volume of preservative solution.
- Prepare slides by placing the resuspended cell sample onto the slide and spreading it evenly.
- Label the slides with appropriate patient identification and date.
- Allow the slides to dry completely before staining.
- Stain the slides using the standardized staining protocol.
- Examine the slides under a microscope and record findings.
- Document all results in the laboratory information system.
