Purpose

To control pre-analytical factors that affect IHC results.

Scope

Fixation to section preparation for IHC.

Responsibilities

Grossing + IHC technicians
Pathologist: interpretation with controls

Materials & equipment

10% NBF, charged slides
Antigen-retrieval reagents

Procedure

Standardise fixation time in 10% NBF; record cold-ischaemia and fixation times.
Prefer EDTA/gentle decalcification for antigen-sensitive targets; limit acid decalcification.
Cut onto charged slides at consistent thickness; dry adequately.
Apply the validated antigen-retrieval method/pH per antibody.
Run positive and negative controls with every batch.
