H&E Staining SOP
Purpose
To produce consistent haematoxylin and eosin staining.
Scope
Manual or automated H&E.
Responsibilities
- Staining technician: run and QC
- Lab manager: reagent schedule
Materials & equipment
- Dewax/xylene, graded alcohols
- Haematoxylin, acid alcohol, bluing reagent, eosin
- Mounting medium, coverslips
Procedure
- Dewax and rehydrate through xylene and graded alcohols to water.
- Stain in haematoxylin for the validated time; rinse.
- Differentiate in acid alcohol with microscope control; blue.
- Counterstain in eosin; dehydrate, clear and mount.
- Check a control slide each run; standardise and log times.

- Staining run log
- Reagent change log
Safety
- Xylene/alcohol flammable and toxic — ventilation and PPE.
