Fluid / Effusion Cytology Processing SOP
Purpose
To outline the standardized procedure for processing fluid and effusion cytology samples to ensure accurate diagnosis.
Scope
This SOP applies to all cytology personnel handling fluid and effusion samples.
Responsibilities
- Cytotechnologist: Perform sample processing and analysis
- Laboratory Technician: Prepare slides and maintain equipment
- Quality Manager: Ensure compliance with SOP and quality control measures
Materials & equipment
- Cytology fluid samples
- Cytology slides
- Fixative solution
- Staining reagents
- Microscope
- Pipettes
- Gloves
- Personal protective equipment (PPE)
Procedure
- Label all samples with patient information and date of collection.
- Centrifuge the fluid sample at 1500 rpm for 10 minutes.
- Decant the supernatant and retain the sediment for processing.
- Resuspend the sediment in an appropriate volume of saline or fixative.
- Prepare slides by placing a drop of the resuspended sediment on a slide and spreading it evenly.
- Fix the slide using the fixative solution for the recommended time before staining.
- Stain the slides using a standard staining protocol appropriate for cytology samples.
- Examine the slides under a microscope and document findings in the laboratory information system.

Safety
