Fixation SOP

Purpose

To standardise fixation so morphology and antigenicity are preserved.

Scope

All specimens requiring formalin fixation.

Responsibilities

  • Grossing technician / pathologist: ensure adequate fixation before processing

Materials & equipment

  • 10% neutral buffered formalin
  • Appropriately sized containers

Procedure

  1. Place tissue in 10% NBF at a fixative-to-tissue ratio of about 10:1.
  2. Slice large/thick specimens (bread-loaf) to allow penetration.
  3. Record time in and out of fixative.
  4. Ensure adequate fixation time appropriate to specimen size before processing.
  5. Minimise cold-ischaemia time; fix promptly after removal.

Safety

  • Handle formalin in a fume hood with PPE; follow the formalin handling SOP.
