Decalcification SOP
Purpose
To outline the procedure for decalcifying bone specimens to ensure optimal histological processing.
Scope
This SOP applies to all personnel involved in specimen handling within the histopathology laboratory.
Responsibilities
- Laboratory Technician: Perform decalcification as per SOP.
- Quality Manager: Ensure compliance with SOP and review records.
Materials & equipment
- Decalcifying solution (e.g., EDTA, hydrochloric acid)
- Specimen containers
- pH meter
- Timer
- Personal protective equipment (gloves, goggles, lab coat)
Procedure
- Label the specimen container with patient information and date.
- Place the bone specimen in the decalcifying solution.
- Monitor the pH of the solution regularly during decalcification.
- Set a timer for the recommended decalcification duration based on specimen size.
- After the designated time, check the specimen for adequate decalcification using a needle or sharp instrument.
- Once decalcified, rinse the specimen thoroughly with running water.
Records
- Decalcification log indicating specimen ID, date, solution used, and duration of decalcification.
Safety
- Always wear appropriate personal protective equipment when handling decalcifying agents.
- Handle decalcifying solutions in a fume hood to avoid inhalation of fumes.
