Antigen Retrieval SOP
Purpose
To outline the procedure for antigen retrieval in immunohistochemistry (IHC) to enhance antibody binding.
Scope
This SOP applies to all personnel performing IHC in the histopathology laboratory.
Responsibilities
- Laboratory Technologist: Perform antigen retrieval procedures
- Quality Manager: Ensure compliance with SOP
- Pathologist: Review and validate results
Materials & equipment
- Citrate buffer
- Tris-EDTA buffer
- Microwave or water bath
- Glass slides with tissue sections
- Pipettes
- Protective gloves
- Safety goggles
Procedure
- Prepare the antigen retrieval buffer according to the protocol
- Deparaffinize tissue sections using xylene and rehydrate in graded alcohols
- Place slides in the antigen retrieval solution in a microwave or water bath
- Heat the solution to the specified temperature for the designated time
- Allow slides to cool to room temperature
- Rinse slides in phosphate-buffered saline (PBS) to remove excess buffer
- Proceed with IHC staining protocol

- Wear protective gloves and goggles during the procedure
- Handle hot equipment with care to avoid burns
