Xylene substitute performing poorly
Also known as: xylene alternative, xylene substitute
The tissue may appear inadequately cleared or exhibit residual staining. This can affect the overall quality of the histological sections.
Where it usually appears: Typically observed during the clearing stage of tissue processing or before embedding in paraffin.
- Improper storage conditions
- Expired or contaminated reagent
- Inadequate volume used
- Incorrect processing times
- Improper temperature during processing
- Check the expiration date of the reagent
- Inspect for cloudiness or particulates in the solution
- Verify the volume used in the processing protocol
- Replace with a fresh batch of xylene substitute
- Adjust processing times according to SOP
- Ensure adequate volume is used for clearing
- Store reagents in a cool, dark place
- Regularly check and rotate stock to use older reagents first
- Train staff on proper handling and usage procedures
