Wax too hot at embedding (melt-back)
Also known as: melt-back, overheating wax
The tissue sections may appear distorted or have a shiny, overly smooth surface. The wax block may show signs of melting or separation from the tissue.
Where it usually appears: Typically observed during the embedding process when the wax temperature is excessively high.
- Wax temperature set too high
- Prolonged exposure of tissue to hot wax
- Improper calibration of embedding center
- Check the temperature setting of the embedding station
- Inspect the wax for signs of excessive melting
- Lower the embedding wax temperature immediately
- Re-embed affected samples at appropriate temperature
- Regularly calibrate embedding equipment
- Monitor wax temperature during embedding process
- Use a thermometer to verify wax temperature
