Uneven differentiation across the slide
The slide shows areas with varying intensity of staining, resulting in some regions appearing overly dark while others are pale.
Where it usually appears: Typically observed during routine H&E staining of tissue sections.
- Inconsistent timing in staining steps
- Uneven application of reagents
- Variations in tissue thickness
- Inadequate washing between steps
- Contaminated reagents
- Verify timing of each staining step
- Inspect for even application of reagents
- Check for consistent tissue section thickness
- Re-stain the slide with consistent timing
- Ensure thorough washing between staining steps
- Re-evaluate reagent quality
- Standardize staining protocols
- Train staff on consistent application techniques
- Regularly calibrate equipment for consistency
