Tubular structures not embedded on cross-section
Tubular structures appear distorted or not visible in cross-section on the slide or block.
Where it usually appears: Typically observed in tissue samples containing glandular or tubular structures, such as kidney or salivary gland.
- Inadequate fixation leading to tissue shrinkage
- Improper orientation of the tissue during embedding
- Insufficient embedding medium penetration
- Incorrect temperature during embedding
- Inadequate trimming of the block
- Verify the orientation of the tissue in the embedding mold
- Check for proper fixation time and conditions
- Inspect the embedding medium for consistency
- Re-embed the tissue with proper orientation
- Adjust the trimming technique to expose tubular structures
- Ensure adequate infiltration of embedding medium
- Follow standardized fixation protocols
- Train staff on proper embedding techniques
- Regularly calibrate embedding equipment
