Tissue shrinkage after processing
Also known as: tissue contraction, shrinkage artifact
Tissue appears smaller than expected; edges may be curled or distorted on the slide.
Where it usually appears: Commonly seen in paraffin-embedded sections during routine histological examination.
- Over-dehydration during processing
- Inadequate embedding medium
- Excessive heat during paraffin infiltration
- Improper fixation time
- Verify processing times and temperatures
- Check for adequate fixation prior to processing
- Reprocess the tissue with adjusted dehydration steps
- Ensure proper embedding medium is used
- Monitor dehydration steps closely
- Use appropriate fixation protocols
- Maintain consistent processing temperatures
