Tissue shrinkage after processing

What it looks like

Tissue appears smaller than expected; edges may be curled or distorted on the slide.

Where it usually appears: Commonly seen in paraffin-embedded sections during routine histological examination.

Likely causes
  • Over-dehydration during processing
  • Inadequate embedding medium
  • Excessive heat during paraffin infiltration
  • Improper fixation time
Immediate checks
  • Verify processing times and temperatures
  • Check for adequate fixation prior to processing
Corrective action
  • Reprocess the tissue with adjusted dehydration steps
  • Ensure proper embedding medium is used
Preventive action
  • Monitor dehydration steps closely
  • Use appropriate fixation protocols
  • Maintain consistent processing temperatures
