Tissue set too deep in the block
Also known as: tissue too deep, embedding depth issue
The tissue appears submerged within the paraffin block, making it difficult to locate on the slide. It may result in incomplete or obscured sections during microtomy.
Where it usually appears: Typically observed during routine embedding of larger specimens or when using excessive embedding medium.
- Inadequate orientation of the tissue during embedding
- Excessive embedding medium used
- Improper cooling or solidification of the block
- Verify the orientation of the tissue in the block
- Check the level of the embedding medium used
- Re-embed the tissue with proper orientation
- Trim the block to expose the tissue adequately
- Ensure proper orientation and positioning of the tissue during embedding
- Use the appropriate amount of embedding medium
