Tissue crumbles away from the wax
Tissue appears fragmented or missing from the paraffin block. Sections may show gaps or voids where the tissue should be present.
Where it usually appears: Commonly observed during sectioning of poorly infiltrated or over-fixed tissue samples.
- Inadequate infiltration of tissue with paraffin wax
- Over-fixation leading to tissue brittleness
- Improper cooling of the paraffin block
- Incorrect microtome settings or blade angle
- Inspect the infiltration time and temperature used for the tissue
- Check the microtome blade for damage or dullness
- Evaluate the cooling process of the paraffin block
- Re-infiltrate the tissue with fresh paraffin wax under optimal conditions
- Adjust microtome settings or replace the blade if necessary
- Use a softer cutting technique or adjust the angle of the blade
- Ensure proper fixation and infiltration protocols are followed
- Regularly inspect and maintain microtome equipment
- Train staff on proper handling of delicate tissues
