Sections vary in thickness across the block
Also known as: thickness variation, inconsistent sections
Sections exhibit uneven thickness, with some areas appearing much thicker or thinner than others. This can lead to poor staining and unclear visualization of tissue structures.
Where it usually appears: Typically observed during routine microtomy when cutting paraffin-embedded tissue blocks.
- inconsistent block orientation
- blade dullness or damage
- incorrect microtome settings
- inadequate embedding
- temperature fluctuations during cutting
- check the microtome blade for sharpness
- inspect the block for proper orientation
- verify microtome settings
- replace or sharpen the microtome blade
- reorient the block properly
- adjust microtome settings for consistent thickness
- regularly maintain and calibrate the microtome
- ensure proper embedding techniques are followed
- monitor temperature during cutting
