Ribbon splits into strips
The tissue ribbon appears fragmented and is not continuous, resembling strips rather than a smooth section.
Where it usually appears: Typically observed during sectioning of paraffin-embedded tissues.
- Blade dullness or damage
- Incorrect microtome settings
- Inadequate tissue fixation
- Improper embedding technique
- Inspect the microtome blade for sharpness
- Verify microtome settings for thickness
- Check for any debris in the cutting area
- Replace or sharpen the microtome blade
- Adjust the microtome settings as necessary
- Re-embed the tissue block if fixation was inadequate
- Regularly check and maintain microtome blades
- Follow proper embedding protocols
- Ensure optimal fixation times and conditions
