Poor rapid-H&E staining on frozen sections
Staining appears faint or uneven with poor differentiation between tissue and background.
Where it usually appears: Commonly observed during the preparation of frozen sections for rapid diagnosis.
- Insufficient fixation time prior to freezing
- Inadequate temperature control during freezing
- Improper section thickness
- Incompatible mounting medium
- Expired or improperly stored reagents
- Verify temperature of the cryostat
- Check fixation time before freezing
- Inspect section thickness
- Re-fix the tissue for the appropriate time
- Adjust cryostat settings for optimal temperature
- Re-stain with fresh reagents
- Standardize fixation protocols
- Regularly calibrate and maintain cryostat equipment
- Ensure reagents are within expiration dates
