PAS background too strong

The slide exhibits a pronounced pink or magenta background staining that obscures the intended structures.
Where it usually appears: Commonly observed in tissue sections with high glycogen content or in poorly processed samples.
- Excessive reagent incubation time
- Inadequate washing steps
- Overconcentration of PAS reagent
- Contamination of reagents
- Verify incubation time with PAS reagent
- Check washing steps for thoroughness
- Reduce incubation time with PAS reagent
- Increase washing time or number of washes
- Dilute PAS reagent if overconcentrated
- Follow SOP for PAS staining precisely
- Use fresh reagents and check for contamination regularly
