Pap stain nuclei too pale

What it looks like

Nuclei appear faint or washed out, lacking the typical blue-purple staining intensity.

Where it usually appears: Commonly observed in cervical cytology specimens during routine Pap staining.

  • Insufficient staining time
  • Inadequate fixation
  • Improper pH of staining solution
  • Degraded or expired reagents
  • Inadequate dehydration prior to staining
Immediate checks
  • Verify the staining time used
  • Check the fixation quality of the slides
  • Assess the pH of the staining solution

  • Re-fix the slides using appropriate fixative
  • Replace any expired reagents
