Pap stain nuclei too pale
Nuclei appear faint or washed out, lacking the typical blue-purple staining intensity.
Where it usually appears: Commonly observed in cervical cytology specimens during routine Pap staining.
- Insufficient staining time
- Inadequate fixation
- Improper pH of staining solution
- Degraded or expired reagents
- Inadequate dehydration prior to staining
- Verify the staining time used
- Check the fixation quality of the slides
- Assess the pH of the staining solution
- Extend the staining time
- Re-fix the slides using appropriate fixative
- Replace any expired reagents
- Regularly calibrate and maintain staining equipment
- Use fresh reagents and monitor their expiration dates
- Implement routine quality control checks on staining procedures
