Only partial sections of the tissue appear
Also known as: incomplete sections, partial cuts
Sections show only fragments of tissue instead of complete slices. The block may appear uneven or damaged.
Where it usually appears: Typically observed during routine microtomy when cutting paraffin-embedded tissue blocks.
- Block is too hard due to improper embedding or over-hardening
- Blade is dull or damaged
- Incorrect cutting angle or speed
- Inadequate support of the tissue block during cutting
- Tissue is not adequately infiltrated with paraffin
- Inspect the microtome blade for sharpness and damage
- Check the embedding medium for proper consistency
- Examine the tissue block for signs of over-hardening
- Replace or sharpen the microtome blade
- Adjust cutting speed and angle
- Re-embed the tissue block if necessary
- Regularly check and replace microtome blades
- Ensure proper embedding techniques are followed
- Maintain consistent paraffin infiltration protocols
