Multiple fragments scattered or not level
Tissue fragments appear unevenly distributed on the slide or within the paraffin block. Some fragments may be partially embedded or protruding from the block.
Where it usually appears: This issue is often noted during the embedding stage of sample preparation, especially with larger or irregularly shaped specimens.
- Inadequate fixation leading to soft tissue
- Improper orientation of the tissue during embedding
- Insufficient embedding medium
- Inconsistent temperature during embedding
- Overloading of the embedding mold
- Inspect the embedding mold for proper orientation and sufficient medium
- Check the temperature of the embedding station
- Re-embed the affected samples ensuring proper orientation and adequate medium
- Use a new embedding mold if the current one is compromised
- Ensure proper fixation protocols are followed
- Use appropriate embedding techniques for different tissue types
- Regularly calibrate and maintain embedding equipment
