The section shows discontinuities or gaps in the cut surface, often appearing as lines or skips in the tissue.
Where it usually appears: Typically observed during the microtomy of hard tissues like bone or calcified specimens.
- Blade dullness or damage
- Incorrect blade angle
- Inadequate tissue fixation
- Improper embedding technique
- Inconsistent microtome settings
- Inspect the blade for chips or dullness
- Verify the blade angle and alignment
- Check the embedding medium for consistency
- Replace or sharpen the blade
- Adjust the blade angle
- Re-embed the tissue if fixation is inadequate
- Calibrate the microtome settings
- Regularly check and maintain blades
- Use appropriate embedding techniques
