GMS (silver) over-stained background
The slide shows a dark, heavily stained background with little contrast to the tissue sections. Silver deposits may obscure cellular details.
Where it usually appears: Typically appears during GMS staining when the staining time is excessive or the developer is too concentrated.
- Prolonged exposure to silver nitrate solution
- Excessive development time in ammoniacal silver solution
- Improper dilution of reagents
- Verify timing of each staining step
- Check the concentration of silver nitrate and developer solutions
- Reduce the staining time
- Dilute the developer solution before use
- Rinse thoroughly to remove excess silver
- Follow the standard operating procedure for GMS staining
- Calibrate timing devices to ensure accurate timing
- Regularly check reagent concentrations
