Frozen sections tear or shred
Sections appear ragged or have uneven edges, with visible tearing or shredding. The tissue architecture is compromised, making it difficult to assess morphology.
Where it usually appears: Typically observed during the cutting of frozen sections, especially with delicate or fatty tissues.
- Improper freezing of the tissue sample
- Dull or damaged microtome blade
- Incorrect section thickness settings
- Inadequate support of the tissue block during cutting
- Excessive thawing of the tissue before sectioning
- Inspect the microtome blade for damage or dullness
- Verify the freezing technique used for the sample
- Check the section thickness settings
- Replace or sharpen the microtome blade
- Re-freeze the tissue sample properly before cutting
- Adjust the section thickness to appropriate levels
- Ensure proper freezing techniques are followed
- Regularly maintain and calibrate the microtome
- Use appropriate embedding media to support delicate tissues
