Counterstain too heavy in IHC
Also known as: overstaining, excessive counterstaining
The tissue sections appear overly dark or muddy, obscuring the specific staining of the target antigen. Details of cellular architecture may be lost due to the heavy background staining.
Where it usually appears: Typically observed in immunohistochemistry (IHC) slides after counterstaining with hematoxylin or similar reagents.
- Excessive incubation time in counterstain solution
- Improper dilution of counterstain
- Inadequate washing after primary or secondary antibody application
- Inappropriate choice of counterstain for the tissue type
- Check the incubation time for counterstain application
- Verify the dilution ratio of the counterstain solution
- Reduce the incubation time in counterstain solution
- Dilute the counterstain more appropriately
- Increase washing time to remove excess counterstain
- Standardize counterstain protocols in SOP
- Train personnel on proper staining techniques
- Regularly calibrate and maintain equipment used for staining
