Condensation / frost on frozen sections
Also known as: frosting, ice crystals
Frost appears as white or cloudy areas on the tissue sections, obscuring cellular details.
Where it usually appears: Usually observed during the sectioning of frozen tissue blocks, particularly in low humidity environments.
- High humidity in the freezing chamber
- Inadequate cooling of the cryostat
- Rapid temperature fluctuations
- Improper handling of the frozen blocks
- Check the temperature settings of the cryostat
- Inspect the humidity levels in the laboratory
- Allow the cryostat to stabilize at the correct temperature
- Ensure proper freezing of the tissue before sectioning
- Maintain consistent cryostat temperature
- Control humidity in the laboratory environment
- Use anti-fogging agents if necessary
