Condensation / frost on frozen sections

What it looks like

Frost appears as white or cloudy areas on the tissue sections, obscuring cellular details.

Where it usually appears: Usually observed during the sectioning of frozen tissue blocks, particularly in low humidity environments.

Likely causes
  • High humidity in the freezing chamber
  • Inadequate cooling of the cryostat
  • Rapid temperature fluctuations
  • Improper handling of the frozen blocks
Immediate checks
  • Check the temperature settings of the cryostat
  • Inspect the humidity levels in the laboratory
Corrective action
  • Allow the cryostat to stabilize at the correct temperature
  • Ensure proper freezing of the tissue before sectioning
Preventive action
  • Maintain consistent cryostat temperature
  • Control humidity in the laboratory environment
  • Use anti-fogging agents if necessary
