Chatter in frozen sections
Also known as: frozen section chatter, frozen artifact
Chatter appears as a wavy or undulating pattern on the tissue section, leading to an unclear image. It may result in areas of loss of cellular detail.
Where it usually appears: Typically seen in frozen sections of delicate tissues or during rapid freezing processes.
- Inadequate freezing of the tissue
- Incorrect microtome settings
- Vibration during sectioning
- Improper handling of the tissue block
- Inspect the microtome for vibrations
- Check the freezing method used
- Examine the tissue block for proper embedding
- Adjust the microtome settings for speed and thickness
- Ensure adequate freezing time before sectioning
- Minimize vibrations in the laboratory environment
- Use proper embedding techniques for delicate tissues
- Calibrate the microtome regularly
- Train staff on optimal sectioning techniques
