Cell loss during liquid-based processing
Also known as: cell loss, sample degradation
Reduced cellularity on the slide, with fewer cells than expected. Possible obscuring of cellular details.
Where it usually appears: Typically observed during the preparation of cytology samples, especially in liquid-based cytology.
- Inadequate sample collection
- Improper fixation
- Insufficient centrifugation
- Excessive washing of the sample
- Old or degraded processing reagents
- Review sample collection technique
- Check fixation time and method
- Examine centrifuge settings
- Repeat sample collection if possible
- Adjust centrifugation speed and time
- Optimize fixation protocol
- Train staff on proper collection techniques
- Regularly calibrate and maintain equipment
- Monitor reagent expiration dates
