Block face will not cut flat (cannot face the block)
The block face appears uneven with visible grooves or ridges, making it difficult to obtain flat sections.
Where it usually appears: Typically seen when preparing paraffin-embedded tissue blocks for microtomy.
- Inadequate embedding of the tissue
- Improper cooling of the block
- Excessive blade wear or dullness
- Incorrect blade angle
- Block temperature too high
- Inspect the blade for sharpness
- Check the embedding medium for proper polymerization
- Assess the temperature of the block
- Replace or sharpen the microtome blade
- Re-embed the tissue if necessary
- Adjust the angle of the blade during cutting
- Regularly maintain and sharpen blades
- Ensure proper embedding techniques are followed
- Store blocks at optimal temperatures
