Background basophilia (blue haze)
The slide exhibits a diffuse blue haze that obscures cellular details, making it difficult to interpret the staining results.
Where it usually appears: Commonly observed in sections with high RNA content or in poorly processed tissues.
- Excessive fixation time leading to over-fixation
- Inadequate dehydration during processing
- Improper pH of the staining solution
- Contamination with basic substances
- Inspect the pH of the staining solutions
- Check fixation time and method
- Review processing steps for dehydration adequacy
- Adjust the pH of the staining solution
- Optimize dehydration steps in tissue processing
- Reduce fixation time if excessive
- Regularly calibrate and maintain pH meters
- Implement standard processing protocols
- Train staff on proper fixation and processing techniques
